Conversion of the Oncogenic 1-Methylguanine 3-Oxide to 3-Hydroxy-1-methylxanthine1

چکیده
tion of 1-methylguanine 3-oxide was not observed (8). This observation, combined with the very low activity of 1-meth ylguanine 3-oxide as a substrate for sulfotransferase in an in vitro assay for activation (10), caused us to compare the metabolic fates of 1-methylguanine 3-oxide and 3-hydroxy 1-methylxanthine with their unmethylated parent 3-hydroxy purines to provide a better understanding on the oncogenic properties of these purine N-oxides.

Conversion of the oncogenic 1-methylguanine 3-oxide to 3-hydroxy-1-methylxanthine.
Deamination of the oncogenic 1-methylguanine 3-oxide occurs to a significant extent in rats to yield 3-hydroxy-1-methylxanthine and its metabolites. When 3-hydroxy-1-methylxanthine is administered, 1-methyl-8-methylthioxanthine can be recovered from urine and released from hepatic protein. No 1-methyl-8-methylthioguanine was detected in urine or bound to protein. There is no evidence of signifi...
متن کاملOccurrence of Methylated Purine Bases in Yeast Ribonucleic Acid*
A number of methylated purine bases have recently been reported to occur in small quantities in the urine of normal human subjects. These include 1-methylguanine, iV*-methylguanine (6-hydroxy-2-methylaminopurine) ,I 1-methylhypoxanthine, and 8-hydroxy-7-methylguanine (1) in addition to 7-methylguanine, previously known as a urinary constituent (2). These compounds were found to be metabolites o...
متن کاملDirect Conversion of Propylene Oxide to 3-Hydroxy Butyric Acid Using a Cobalt Carbonyl Ionic Liquid Catalyst
The reported catalytic system demonstrates the possibility of efficient mass production of 3-hydroxybutyric acid (3-HBA) from inexpensive raw materials. The direct coupling of propylene oxide, water, and CO was catalyzed by 1-butyl-3-methylimidazolium cobalt tetracarbonyl ([Bmim][Co(CO)4]) ionic liquid to form 3-HBA with >99% conversion (49% selectivity) under mild conditions.

متن کامل4-Hydroxy-1-(3-pyridyl)-1-butanone, an indicator for 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone-induced DNA damage, is not detected in human pancreatic tissue.
Tobacco smoking is the only known etiologic agent that causes pancreatic cancer. The tobacco-specific nitrosamine 4-(methylnitrosamino)-I-(3-pyridyl)-1-butanone (NNK) is a potent carcinogen in laboratory rodents that, independent of the route of administration, induces primarily lung adenocarcinoma (1). When administered in drinking water, NNK and its metabolite 4-(methy1nitrosamino)-1(3-pyridy...
